基于微流控技術(shù)的腸上皮屏障功能檢測(cè)系統(tǒng)

由于疾病引起或藥物毒性導(dǎo)致的上皮屏障功能障礙可能危及生命。上皮屏障破壞主要表現(xiàn)為上皮細(xì)胞的細(xì)胞通透性增加。上皮屏障的細(xì)胞通透性的體外測(cè)試通常通過(guò)在剛性膜上培養(yǎng)細(xì)胞來(lái)實(shí)現(xiàn),這種傳統(tǒng)的Transwell系統(tǒng)不適合高分辨率動(dòng)力學(xué)測(cè)量,不能像3D培養(yǎng)一樣支撐更多的模擬和研究。微流控技術(shù)在體外模擬領(lǐng)域迅速發(fā)展。微流控技術(shù)固有的小尺寸特點(diǎn)使其恰好能與管狀上皮連接,以便通過(guò)灌注提供剪切應(yīng)力和連續(xù)培養(yǎng)基。典型的微流體溶液利用人造膜進(jìn)入上皮細(xì)胞而不通過(guò)ECMECM是參與分化和上皮-間充質(zhì)轉(zhuǎn)化的細(xì)胞信號(hào)轉(zhuǎn)導(dǎo)中的關(guān)鍵參數(shù)。 

基于此,荷蘭Mimetas公司聯(lián)合創(chuàng)始人兼總經(jīng)理Paul Vulto博士及其團(tuán)隊(duì)開(kāi)發(fā)了一種腸上皮屏障功能檢測(cè)系統(tǒng)來(lái)培養(yǎng)灌流的ECM支持的上皮細(xì)胞并以無(wú)膜方式測(cè)試它們的屏障功能。作為一個(gè)例子,研究者們開(kāi)發(fā)了一種顯示細(xì)胞極化、緊密連接形成和關(guān)鍵受體表達(dá)的腸道上皮細(xì)胞模型。 40個(gè)腸道模型在器官芯片平臺(tái)(OrganoPlate)中以管狀形式生長(zhǎng),可從頂端和基底兩側(cè)進(jìn)入。
