微流控技術制備微血管并實現血管新生

組織工程的最終目標是制造人體器官。然而,在這之前必須重建和表征三維組織。目前無法制造可操作的脈管系統,這限制了組織工程的發展。基于此,美國海軍研究實驗室Andre A. Adams和北卡羅萊納州立大學Michael A. Daniele團隊使用微流體技術制造了獨立的小直徑血管,其具有可重現正常生物功能的組織細胞層。隨著培養時間的推移,內皮細胞在腔壁上形成單層結構并分泌細胞外基質。在整合到含有成纖維細胞的三維凝膠中后,微血管延伸并形成中空分支,其與相鄰的毛細血管吻合成網。人工制造的微血管以及延伸出來的血管都支持流體和顆粒的灌注。以上所述微血管可以被設計為不同的直徑,并且可完成血管新生和融合,這將成為組織工程血管形成極具價值的工具。

該研究的目的是開發一種有效的人體血管構建方法,這些制造的血管將通過血管生成,管腺增生和融合的自然過程來完成血管原始網絡的形成(圖1)。通過紫外(UV)光交聯聚乙二醇(PEG)和甲基丙烯酸明膠(GelMA)制備人內皮微血管(HEMV),其可經歷新血管生成以及與正常人血管成熟相關的其他發育過程。
